Development & Validation of Pharmacological Tools for the Preclinical Testingof Anticoagulants

(eng) Despite the clinical use of well-established and newer anticoagulant drugs, venous thromboembolism (VTE) remains a major public health problem associated with high morbidity and mortality. The search for new anticoagulant drugs with an improved benefit/risk ratio is therefore a main challenge for the pharmaceutical industries. Success in this endeavour is driven by the right completion of the preclinical studies. In this work, a series of relevant and reliable pharmacological tools modeling thrombotic disorders was successfully developed and validated to evaluate prospective anticoagulant compounds. This preclinical platform consisted in three drug testing levels: (1) in vitro enzymatic assays on isolated coagulation factors (i.e. thrombin, factor Xa, tissue factor/factor VIIa complex and factor XIIa); (2) functional ex vivo thrombin generation assays in human plasma and (3) an in vivo venous thrombosis model induced by stasis and endothelial damages and coupled to thrombin generation assay in the rat. This preclinical testing platform was developed and evaluated during the evaluation of chemically distinct compounds selected for their potential anticoagulant and antithrombotic properties. Three types of products were investigated in order to identify promising innovative anticoagulants for the prevention and treatment of VTE: coumarins, the tick protein Ixodes ricinus contact phase inhibitor (Ir-CPI) and leaf extracts/fractions from Croton zambesicus.

(fre) Malgré l’utilisation de thérapies anticoagulantes conventionnelles ou novatrices, les pathologies thromboemboliques veineuses restent un problème de santé publique majeur associé à une morbidité et une mortalité élevées. La recherche de nouveaux agents anticoagulants est donc un défi important pour l’industrie pharmaceutique. L’accomplissement fructueux de cette mission est guidé par la bonne réalisation des études précliniques. Dans ce travail, une série d’outils pharmacologiques pertinents et fiables comme modèle de troubles thrombotiques ont été développés et validés avec succès afin d’évaluer de potentiels composés anticoagulants. Cette plateforme préclinique s’est articulée en trois niveaux de testing des drogues : (1) des tests enzymatiques in vitro sur facteurs de coagulation isolés (c. à d. la thrombine, le facteur Xa, le complexe FT/FVIIa et le facteur XIIa) ; (2) des tests fonctionnels ex vivo de génération de thrombine dans le plasma humain et (3) un modèle in vivo de thrombose veineuse induite par la stase et par lésion de l’endothélium combiné à des tests de génération de thrombine dans le plasma chez le rat. Cette plateforme de tests précliniques a été développée et testée durant l’évaluation de composés chimiquement distincts ayant été sélectionnés pour leurs propriétés anticoagulantes et antithrombotiques potentielles. Trois types de produits ont été investigués dans le but d’identifier d’éventuels agents anticoagulants originaux pour la prévention et le traitement des troubles thromboemboliques veineux : les coumarines, la protéine de tique Ixodes ricinus contact phase inhibitor (Ir-CPI) et des extraits/fractions de feuilles de Croton zambesicus.
